Bright Red Dot on Gold Buffalo Coin.?

I have a 2006 Proof Gold Buffalo never been touched by human hands but is has a Bright red dot on the Indians cheek. I need to know if that makes it worth more or less?

Regardless of the cause, a red dot would not increase its value. It sounds as if the coin is either in a thin sealed mylar mint packaging, or a hard "plastic" case. It is possible the red dot is attached to its container. If it is attached to the coin, it could increase in size over the years depending upon what it is. I would recommend taking the coin to a local coin expert or two for an evaluation. I hope that helps.
